Semax (Met-Glu-His-Phe-Pro-Gly-Pro) is a synthetic ACTH(4-10) analogue. In Russian preclinical work it has been reported to modulate neurotrophins, including BDNF and TrkB signalling (Dolotov et al., 2006), with effects on NGF described in later Russian preclinical studies, and to show neuroprotective and cognitive effects in animal models. Neurotrophin modulation is one proposed mechanism, not an established primary mode of action.

Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a synthetic tuftsin analogue studied for anxiety-related, GABA-system, monoamine and immune endpoints. The relevant evidence includes rat and mouse gene-expression, neurochemical and behavioral studies plus preclinical antiviral models; it does not establish clinical effects or any effect of the blend.

Both peptides contain a C-terminal Pro-Gly-Pro motif associated with increased resistance to proteolytic degradation in this peptide family; it does not ensure complete enzymatic stability. This is a common design principle originating from the same research group.

Why the Combination?

Research Rationale

The rationale for the Semax/Selank Mix is based on the recognition that cognitive performance and emotional regulation are inseparably linked. Anxiety and stress impair cognitive function, and conversely, cognitive overload can amplify anxiety. The research rationale is to examine cognitive and anxiety-related endpoints in the same experimental system. This does not establish that the blend affects either endpoint or that its effects are complementary, additive or synergistic:

Semax - cognitive axis:

  • Produced strain- and region-dependent changes in monoamine levels and metabolites in mice; these data do not establish mood stabilization
  • Changed immune-gene expression in mouse spleen and showed antiviral activity in preclinical influenza models; these findings do not establish clinical effects or effects of the blend

Scientific Evidence

Panikratova et al. (2020, Doklady Biological Sciences, PMID 32342318): This study examined resting-state functional connectivity in healthy subjects who received Semax, Selank or placebo as SEPARATE administrations, not as a combination. It reported distinct connectivity effects for each peptide on predefined regions of interest including the amygdala. It does not test the two peptides given together, so it is not evidence about the mix.

Vasil'eva et al. (2020, Neurochemical Journal): This Russian preclinical study compared Selank, Semax and Noopept given SEPARATELY by different routes. In BALB/c mice, route-dependent nootropic and anxiolytic effects were reported; C57BL/6 mice largely did not show the same effects, and one comparison reported an opposing Semax anxiety result. The strain and route dependence, and the fact that the peptides were tested separately, mean this does not establish how a Semax/Selank mix behaves.

Mechanistic Findings Reported Separately

No published study has tested the Semax/Selank combination itself. The pathways below are reported SEPARATELY for each peptide, mostly in Russian preclinical work; their combined activity has not been tested, so this is a map of the individual mechanisms, not a demonstrated five-system combination effect:

  1. Neurotrophin-related findings (Semax): Rat studies report time- and region-dependent changes in BDNF, NGF and related receptors. Dolotov et al. (2006) supports BDNF/TrkB-related effects, while later preclinical work addresses NGF. Neurotrophin modulation is one proposed mechanism, not an established primary mode of action.

  2. GABAergic findings (Selank): Rat gene-expression and in-vitro binding studies support GABA-system modulation as a proposed mechanism. They do not establish enhanced inhibitory neurotransmission, absence of sedation or the behavior of the blend.

  3. Monoaminergic System (both): Semax influences dopaminergic pathways, Selank modulates serotonin and noradrenaline metabolism. No study has shown broader monoaminergic modulation from combined administration.

  4. NO-related findings (Semax): In a Wistar-rat incomplete-global-ischemia model, Semax prevented excess cortical NO production and neurological disturbances. This association does not establish NO modulation as the causal neuroprotective mechanism or a general effect in oxidative stress.

  5. Immune-related findings (Selank): In mice, Selank and its fragments changed expression of chemokine, cytokine and receptor genes in spleen; separate preclinical studies reported antiviral activity. These findings do not establish phagocyte activation by Selank, clinical immunomodulation or an effect of the blend.
